Technologies

The slogan Make genome editing easier represents the goal of our partner. Ubigene Biosciences was able to develop a genome editing platform based on the CRISPR/Cas9 technology. These are the new improved technologies CRISPR-U™ and CRISPR-B™. Both new technologies enable easy achievement of knockout (KO), point mutation (PM) and knock-in (AI) in vitro and in vivo. Different types of gene editing vectors are available for different species such as mouse, rat and human. Furthermore, there is a virus packaging service that includes lentiviruses, adenoviruses, and adeno-associated viruses, among others.

CRISPR-U™ technology features improved efficiency in double-strand breakage and homologous recombination. CRISPR-B™ is improved over conventional CRISPR/Cas9 technology in terms of efficiency and precision. CRISPR-B™ technology is used for gene editing of bacteria and fungi.

The cell biology platform is specialized for primary cells. There are over 400 different types of primary cells available. The platform provides culture strategies for these, as well as related products fr the respective cell types. In addition, services such as cell isolation, extraction and validation are provided.

Zebrafish Research

Zebrafish as model organism

Zebrafish serve as model organisms for numerous reasons. They provide the opportunity for holistic animal studies, allowing complex biological processes to be simulated. Their small size and low husbandry requirements combined with a high reproductive rate make them suitable for low-cost, high-throughput research on organisms. The entire genome is sequenced and shows high concordance with the human genome. The gene editing tools are already very sophisticated.

Another advantage of zebrafish as a model organism is that its embryos are large and transparent, making observational studies very easy. Their development is very rapid in vivo and in vitro (a few days). Ethical risks are very low with zebrafish, which allows research projects to be carried out quickly.

Transgenic zebrafish

The Tol2 transposon system can be used to generate transgenic fish. The system creates high integration efficiency and short processing time. The system consists of the Tol2 transposons and the transposase. The transposase plasmid is transcribed into mRNA in vivo, and injected into the zebrafish zygote along with the expression plasmid. The transposase excises the sequence between the Pol2 transposons and integrates it into the zebrafish genome of the zygote. Gene typing can be used to monitor the successful integration of the target gene and select the appropriate individuals. The system is used for drug screening, disease control and gene function studies.

Knockout zebrafish

To produce knockout zebrafish, the CRISPR/Cas9 method is used. This method is suitable for a wide range of gene modifications of almost all cell types. The principle of CRISPR/Cas9 is based on the binding by base pairing of crRNA (CRISPR-derived RNA) with tracRNA (trans-activating RNA). The tracrRNA/crRNA complex (alternatively, a synthetic gRNA can be used) binds the despiralized target DNA, causing the Cas9 nuclease enzyme to cut the double-stranded target DNA, adjacent to the "proto-spacer adjacent motif," abbreviated PAM. This results in a sequence-specific double-strand break. The double-strand break is repaired by cellular DNA repair mechanisms in which DNA sequence fragments are added, removed or replaced by homologous direct repair (HDR) or non-homologous end joining (NHEJ).

Knockout zebrafish are used for drug screening, disease models and gene function studies.
