The properties of chromatography resins

Since there are different types of resins, both for analytical and purification purposes, there are correspondingly different polymers with different modifications.

The special feature of chromatography resins is their flexibility, as the columns themselves can be packed. Depending on the application goal, resins can be exchanged directly, for example to avoid contamination in pharmaceutical analyses, or reused for purification. The scale is also very flexible as a result.

The physical and chemical properties of the analytes, such as proteins or nucleic acids, are very different. Consequently, these differences, for example in size and polarity, can be used for chromatography, because these properties determine, among other things, the choice of resins.

The different types of chromatography

Immobilized metal affinity chromatography (IMAC)

Are you looking for IMAC resins for rapid separation in the lab or for preparative process-oriented applications? Then Marvelgent Biosciences is the right place for you, as our partner company offers many different formats of IMAC resins.

Available are the nickel ion-based resins, Ni-NTA agarose and Ni-NTA agarose 6FF resins, which efficiently purify histidine-tagged proteins with high yield.

There are also the Ni-PentaTM resins. These have highly selective affinity for His-tagged proteins, through a proprietary pentadentate ligand. Furthermore, this provides impressive stability on the Matix surface, which is unaffected by a variety of chemicals, EDTA and DDT. Thus, direct separation of target proteins in various buffers, including culture supernatants, is possible. In addition, leaching of toxic nickel ions is negligible, which is protective for the target protein.

Furthermore, for higher affinity to the His-tag, cobalt ion pre-loaded resins can be used, which lead to higher product purity. The Co-NTA resins are also of interest to avoid the toxicity of the nickel ions that leak out.

Product selection from Marvelgent Biosciences

There are different solutions for different difficulties of IMAC. Therefore, we would like to present a selection of solutions with product suggestions below:

  • Initial situation: Your analytes have post-translational modifications
  • Difficulty: modifications may decrease binding affinity to metal ions, as the His tag may not be optimally accessible
  • Solution: IMAC resins with higher binding capacity, e.g. Ni-IDA or Ni-NTA resins.

  • Initial situation: Their analytes contain cysteine (SH binding), they contain cysteine and are metal ion reactive, their analytes are oxidation sensitive, susceptible to proteolysis by metalloproteases or imidazole sensitive.
  • Difficulty: In most cases, the presence of DTT is required. In some cases, however, ß-ME or EDTA is also needed. Furthermore, a large amount of imidazoles cannot be used with sensitive analytes to compete with His-tags for nickel ions, but minimal use is required.
  • Solution:For all the difficulties described, the use of Ni-PentaTM resin is possible. The resin exhibits tolerance to DTT, ß-ME and EDTA. For the case of imidazole sensitivity, Ni-PentaTM resin can also be used, but Co-NTA resin can also be used.
