Affinity chromatography is an important instrument for the purification of biological molecules such as enzymes, antibodies or receptor proteins. The principle of affinity purification is based on the specific interactions between the various molecules, such as the interaction of antibody and antigen. The conventional process for protein purification is usually labor intensive and yields are often very low. On the other hand, affinity chromatography is very specific and takes significantly less time. The purified protein is of high quality with high yield and high specific activity.

The most widely used affinity matrix is based on Sepharose (pearl shape of the cross-linked porous structure of agarose). Separopore® from bioWORLD corresponds to Sepharose in all physical and chemical properties. The Separopore™ Affin chromatography combines size fractionation and the binding of the target molecules to the Affin matrix. This is followed by the release of the molecule from the matrix using various reagents.

Streptavidin Separopore® 4B from bioWORLD effectively uses the strong interactions between biotin and straptavadin or the somewhat weaker interactions between 2-iminobiotin and streptavidin. It can be used to purify antigens by incubating biotinylated antibodies with antigens. The biotinylated antibody-antigen complex binds to Streptavidin Separopore, from which the antigen can be eluted. The immobilized streptavidin binds biotin and biotinylated substances and can be used for affinity chromatography applications. The basic matrix is a rigid, highly cross-linked pearl agarose with high chemical stability.
