The most critical factor for successful staining is choosing an antibody that specifically binds to the target antigen. Indirect detection requires a labeled secondary antibody and, in some cases, a third layer of signal amplification to detect the antigen-primary antibody complex. Compared to directly labeled primary antibodies, the indirect detection is more sensitive and more important for the effective identification of antigens with low frequency, rare epitopes and under conditions of a suboptimal antigen-antibody binding.
