ArkAb - Human Monoclonal Chimeric Antibodies

ArkAb has been set up in 2018 as a subsidiary of B Cell Design. ArkAb develops raw materials for research in health and in vitro diagnostic market. The French company specializes in human chimeric monoclonal antibodies (human constant region of the heavy chain) for research or further manufacturing use. ArkAb offers catalog products as well as custom made development services.

Today, manufacturers of immunoassays for in vitro diagnostics need antigens and positive controls for their kits on the market. Most of the laboratories use human blood from sick patients, called disease state plasma, as positive controls, quality control and standards for their assays. Because all patients are different, human plasma cannot be reproducible from batch to batch, which implies new calibrations of all the tests. In addition to obvious ethical problems, some parameters are difficult to source, and so they are subject to speculation with important price variations. To provide solutions to these problems, ArkAb has developed a catalog of industrial and reproducible products manufactured by a patented process based on genetically modified mice. These mice are engineered in a way that their cells mostly produce one isotype of specific antibodies. These antibodies are already humanized enough to be recognized as human antibodies! Within a few weeks, ArkAb obtains a master cell bank that allows an in vitro industrial robust production of the antibody of interest. This production is fully reproducible and can be used year after year. Other humanization techniques are long, blind projects as they need to pass all the steps to know whether the expected antibody with human features can be produced. In contrast, ArkAb's approach drastically decreases the delays and costs for production of chimeric monoclonal antibodies, increasing the success rate of having a functional humanized antibody with human features. ArkAb products are a reliable alternative to disease state plasma. It provides manufacturers of immunoassays for in vitro diagnostics a reproducible, secured and sustainable source of monoclonal chimeric antibodies as “synthetic” plasma for their quality controls.

ArkAb monoclonal chimeric antibodies are used in the fields of Anti-infectious diseases, autoimmunity, allergies and food intolerance. The insertion of a human cassette carrying the gene of the immunoglobulin heavy chain constant region in the endogenous locus leads to the expression of human chimeric antibodies directly by the mouse lymphocytes. Therefore, these mice are secreting antibodies with the following properties: Only one human isotype per mouse line, the heavy chain constant region is human, the variable regions are from the mouse and the mouse's complete immune repertoire is maintained.
