Aimplex Biosciences Inc. is our partner for customized and premixed bead-based multiplex immunoassay panels. These are designed for the use with standard flow cytometers, eliminating the need for expensive specialty equipment.

AimPlex Biosciences, formerly YSL Bioprocess Development Co, is headquartered in Pomona, California, USA. Since its founding in 2011, ‘Golden State’ scientists have been developing comprehensive solutions for biomedical testing using multiplex bead-based immunoassays. All immunoassays use existing analytical platforms of flow cytometry for highly sensitive detection. This offers researchers who often have limited budgets the advantage of setting up special further analysis equipment.

The overall goal is to develop and deliver high quality and cost effective flow cytometry assay reagents. Currently, approximately 400 immunoassays are offered in the rapidly evolving product portfolio.

How does AimPlex Multiplex Assay Technology work?

The AimPlex Multiplex Assay Technology utilizes different populations of beads. They can be divided according to size and fluorescence intensity. In size there are two gradations; 4 and 5 microns (4S and 5S). There are a total of 12 levels of fluorescence intensity. As a result, the technology of AimPlex Biosciences can detect up to 24 analytes simultaneously. This happens in a single reaction with a sample of only 15μl.

The bead populations are then determined using a standard flow cytometer. This requires a laser with either blue 488 nm and a red 635 nm or just a 488 nm laser. The maximum emission of the bead classification dye is 700 nm.

The method is similar to the sandwich ELISA assay. Each population of beads is conjugated to a specific antibody which captures the desired protein in the sample like a cytokine. The amount of analyte captured is detected via a biotinylated antibody against a secondary epitope of the protein, followed by streptavidin-R-phycoerythrin treatment. The fluorescence intensity of R-phycoerythrin on the bead is then measured on a conventional flow cytometer. Conclusions on the protein concentration in the sample can then be easily concluded by comparison with the standard curve of the fluorescence signals. The latter is obtained by serial dilution of a known concentration of the protein to be examined.
