It inhibits B-Raf (wildtype), BRAFV600E and C-Raf with IC50s of 8.3 nM, 2.4 nM and 1.4 nM in cell-free assays, respectively. TAK-632 shows potent anti-proliferative activity in vitro against melanoma cell lines A375 and SK-MEL-2. It shows high in vivo antitumor efficacy in mouse model with SK-MEL-2 subcutaneous xenograft.
