Epidermal Growth Factor
Epidermal growth factor (EGF) is a protein that stimulates the growth of various epidermal and epithelial tissues1. It consists of 54 amino acids (6.3 kDa), which are linked by 3 intramolecular disulfide bonds2. It occurs increasingly in the initiation of mitosis. For research purposes it can be used both in vitro and in vivo. In some fibroblasts it also stimulates the described growth in cell culture1.

Epidermal growth factor receptor
EGF is one of the major ligands of the epidermal growth factor receptor family (EGFR). The receptor belongs to the class of transmembrane glycoproteins and consists of an extracellular receptor domain, a transmembrane region and an intracellular domain which functions as a tyrosine kinase3. When EGF binds to EGFR, autophosphorylation of the receptor tyrosine kinase takes place, which leads to the activation of certain signal transduction pathways. Among other pathways, cell proliferation and cell differentiation are regulated by this type of signal transduction 4.

Research areas
EGFR plays a special role in the study of cancer, as it is overexpressed in a variety of tumor cells. Overexpression of EGFR has been found particularly in solid tumors, e.g. Breast cancer, head and neck cancer, non-small cell lung cancer (NSCLC), kidney cancer, ovarian cancer and colon cancer 5. The activation of the receptor is also associated with resistance to chemotherapy and radiation. The described properties of EGFR make the receptor an important therapeutic target in the treatment of cancer, since interactions with EGFR may lead to an interruption of signal transduction 6.
Particularly inhibitors of EGFR were monoclonal antibodies directed against the extracellular receptor domain and low molecular weight compounds7. Both monoclonal antibodies and low molecular weight compounds can cause cell proliferation to be disrupted and apoptosis facilitated. Consequently, the cytotoxic as well as cytostatic properties can be attributed to the substances mentioned 7.
