Specificity:
EMR3
Clone:
3D7
Immunogen:
ARHO-EMR3-CD97 (EGF1) transfectants.
Fusion Partner:
Spleen cells from immunised Armenian hamster were fused with cells of the mouse SP2/0 myeloma cell line.
Specificity Note:
This product recognises human epidermal growth factor (EGF) module-containing mucin-like hormone receptor 3 (EMR3) which is a 56kD member of the EGF-7 transmembrane (TM7) family of adhesion receptors. EMR3 is expressed at the cell surface as a heterodimer. The molecule is predominantly expressed on granulocytes and at lower levels on mature myeloid cells monocytes and dendritic cells. EMR3 is absent on lymphocytes haematopoietic stem cells and myeloid progenitors. Studies suggest that the EMR3 molecule is up-regulated during late stages of neutrophil differentiation and is a marker for terminally differentiated cells. The exact functions of EMR3 and its ligands have not yet been determined.
Preparation:
Purified IgG prepared by affinity chromatography on Protein G from tissue culture supernatant
Buffer Solution:
Phosphate buffered saline pH7. 4
Preservative Stabilisers:
0. 09%Sodium AzideSuggested
Flow Cytometry:
Use 10ul of the suggested working dilution to label 106 cells in 100ul. Suggested
Dilution:
Flow Cytometry - 1/50 - 1/200
Function:
Receptor probably involved in myeloid interactions during immune and inflammatory responses. A ligand for the soluble form of this receptor is present at the surface of momocytes-derived macrophages and activated neutrophils.
Subunit:
Forms a heterodimer consisting of a large extracellular region (alpha subunit) non-covalently linked to a seven-transmembrane moiety (beta subunit) (By similarity).
Subcellular Location:
Cell membrane; Multi-pass membrane protein.
Subcellular Location:
EGF-like module-containing mucin-like hormone receptor-like 3 subunit alpha: Secreted extracellular space. Note=A soluble form is also detected (alpha subunit).
Tissue Specificity:
Displays a predominantly leukocyte-restricted expression with highest levels in neutrophils monocytes and macrophages.
Ptm:
Proteolytically cleaved into 2 subunits an extracellular alpha subunit and a seven-transmembrane subunit (By similarity).
Similarity:
Belongs to the G-protein coupled receptor 2 family. LN-TM7 subfamily [view classification].
Similarity:
Contains 2 EGF-like domains.
Similarity:
Contains 1 GPS domain. 1. Stacey. M. et al. (2001) Human epidermal growth factor (EGF) module-containg mucin-like hormone receptor 3 is a new member of the EGF-TM7 family that recognises a ligand on human macrophages and activated neutrophils. 2. Matmati. M. et al. (2007) The human EGF-TM7 receptor EMR3 is a marker for mature granulocytes.
