Clone:
MEM-18
Fusion Partner:
Spleen cells from mice immunised BALB/c mice were fused with cells from the NS1-Ag4/1 mouse myeloma line.
Specificity Note:
This product antibody recognises a monocyte surface antigen of 55 kD classified as CD14. The CD14 molecule is found predominantly on monocytes and macrophages in flow cytometry it is less strongly expressed on granulocytes and is absent from stem cells and myeloid cells of very early differentiation states. In immunohistology the CD14 molecule is found to be present on Langerhans cells follicular dendritic cells histocytes and high endothelial venules. In ELISA clone MEM-18 recognises the soluble form CD14. Clone MEM-18 is reported to block the binding of bacterial lipopolysaccharide (LPS) to monocytes (2). Removal of sodium azide is recommended prior to use in functional assays.
Preparation:
Purified IgG prepared by DEAE - Chromatogaphy
Buffer Solution:
Phosphate buffered saline
Preservative Stabilisers:
0. 09%Sodium AzideSuggested
Flow Cytometry:
Use 10ul of the suggested working dilution to label 106 cells in 100ul. Suggested
Dilution:
Western Blotting - Non-reducing conditions
Function:
Accessory subunit of the mitochondrial membrane respiratory chain NADH dehydrogenase (Complex I) that is believed to be not involved in catalysis. Complex I functions in the transfer of electrons from NADH to the respiratory chain. The immediate electron acceptor for the enzyme is believed to be ubiquinone.
Subunit:
Complex I is composed of 45 different subunits.
Subcellular Location:
Mitochondrion inner membrane; Matrix side. 1. Pltz. S. G. et al. (2001) The interaction of human peripheral blood eosinophils with bacterial lipopolysacchride is CD14 dependent.
