Localization:
Secreted Trypsin is a 24 kDa enzyme of the serine proteinase family. It is produced in the pancreas as an inactive precursor trypsinogen but the active enzyme is located in the gastrointestinal tract where it degrades proteins to large peptides. Trypsin prefers lysine and arginine residues. One of the substrates for trypsin is chymotrypsinogen which is cleaved to produce active chymotrypsin. High levels of immunoreactive trypsin in the bloodstream can indicate pancreatic malfunction and this indicator is used to screen for and diagnose Cystic Fibrosis.
Function:
Has activity against the synthetic substrates Boc-Phe-Ser-Arg-Mec Boc-Leu-Thr-Arg-Mec Boc-Gln-Ala-Arg-Mec and Boc-Val-Pro-Arg-Mec. The single-chain form is more active than the two-chain form against all of these substrates.

Defects in PRSS1 are a cause of hereditary pancreatitis (HPC) [MIM:167800]; also known as chronic pancreatitis (CP). HPC is an autosomal dominant disease characterized by the presence of calculi in pancreatic ducts. It causes severe abdominal pain attacks.
