Mouse
Specificity:
This antibody reacts with neuroendocrine neoplasms of neural as well as epithelial types. This antibody is specific to a 38 kD protein.
Positive Control:
Pancreas
Target:
Synaptophysin
Localization:
Integral membrane protein. Synaptic vesicles.
Concentration:
tissue culture supernatant Storage
Buffer:
Tissue culture supernatant containing 0. 09% sodium azide
Application Note:
This antibody may be diluted to a titer of 1:25-1:50 in an ABC method. However depending on the staining conditions employed we suggest that final dilution should be determined by the user. Cellular
Localization:
Integral membrane protein. Synaptic vesicles. Synaptophysin is a 38kDa glycoprotein present in the membrane of neuronal presynaptic vesicles in brain spinal cord retina vesicles of adrenal medulla neuromuscular junctions and endocrine cells. It is also expressed by neuroendocrine cells throughout the body both normal and neoplastic. Synaptophysin is a useful marker for the identification of normal neuroendocrine cells and neuroendocrine neoplasms. This antibody reacts with neuroendocrine cells of human adrenal medulla carotid body skin pituitary gland thyroid lung pancreas and gastointestinal mucosa. It also reacts with a wide spectrum of neuroendocrine neoplasms of neural type including neuroblastomas ganglioneuroblastomas ganglioneuromas pheochromocytomas chromaffin and non-chromaffin paragangliomas.
Function:
Possibly involved in structural functions as organizing other membrane components or in targeting the vesicles to the plasma membrane.
Subunit:
Homohexamer or homotetramer.
Subcellular Location:
Cytoplasmic vesicle secretory vesicle synaptic vesicle membrane; Multi-pass membrane protein. Cell junction synapse synaptosome.
Tissue Specificity:
Characteristic of a type of small (30-80 nm) neurosecretory vesicles including presynaptic vesicles but also vesicles of various neuroendocrine cells of both neuronal and epithelial phenotype.
Domain:
The calcium-binding activity is thought to be localized in the cytoplasmic tail of the protein.
Ptm:
Ubiquitinated; mediated by SIAH1 or SIAH2 and leading to its subsequent proteasomal degradation (By similarity).
Similarity:
Belongs to the synaptophysin/synaptobrevin family.
Similarity:
Contains 1 MARVEL domain.
