Immunogen:
Synthetic peptide corresponding to a region of human MLH1 encoded within exon 20.
Specificity Note:
This product is specific for human MLH1 (Homologue 1 of E. coli MutL Protein). MLH1 is housekeeping gene which plays an important role in DNA mismatch repair. In humans the protein encoded by MLH1 forms at least two dimeric factors with either PMS2 or MLH3. These heterodimers are reported to form larger complexes made up of several other DNA repair proteins including MSH2 and MSH6. In addition to DNA mismatch repair MLH1 is thought to pay a role in the induction of apoptosis in response to DNA damage. Mutations in MLH1 have been implicated in certain types of colon cancer.
Immunohistology:
This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6. 0 is recommended for this purpose. Histology: Normal colon

Antisera to human MLH1 were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG was prepared from whole serum by affinity chromatography.

It is a human homolog of the E. coli DNA mismatch repair gene mutL consistent with the characteristic alterations in microsatellite sequences (RER+ phenotype) found in HNPCC. Alternatively spliced transcript variants encoding different isoforms have been described but their full-length natures have not been determined.
