Genway ID: |
GWB-EB0FA5 |
Isotype: |
IgG |
Immunogen: |
Synthetic peptide (Human). |
Antigen Species: |
Human |
Specificity: |
This product recognises phosphorylated STAT5. |
Positive Control: |
3T3 L1 adipocytes +/- Leukemia Inhibitory Factor (LIF) |
Target: |
STAT5 |
Localization: |
Nuclear |
Purification Note: |
Purified from rabbit serum by sequential epitope-specific chromatography. The antibody has been negatively preadsorbed using a non-phosphopeptide corresponding to the site of phosphorylation to remove antibody that is reactive with non-phosphorylated STAT5. The final product is generated by affinity chromatography using a STAT5-derived peptide that is phosphorylated at tyrosine 694. Storage |
Buffer: |
PBS pH 7. 3 (without Mg2+ and Ca2+) containing 50% glycerol 1. 0mg/ml BSA (IgG protease free). and 0. 05% sodium azide. |
Application Note: |
WB: Use at a dilution of 1/1000. Detects a band of approximately 95 kDa (predicted |
Molecular Weight: |
90. 6 kDa). Can be blocked with Phospho and non phospho peptide pair for STAT5a [pY694] Not tested in other applications. Optimal dilutions/concentrations should be determined by the end user. Cellular |
Localization: |
Nuclear STAT5a and STAT5b knockout mice show important roles in prolactin-mediated mammary gland development and growth hormone-mediated induction of sexual dimorphism respectively. Abnormal activity of certain STAT family members particularly STAT3 and STAT5 is associated with a wide variety of human malignancies including hematologic breast head and neck and prostate cancers. STAT5A and STAT6 are selectively activated when the heart is subjected to ischemic injury whereas activation of STAT3 and STAT5A is involved in myocardial hypertrophy. STAT5 and STAT3 as these two STAT molecules are required for normal breast development and involution respectively and may play an important role in breast carcinogenesis. The activation of STAT5 proteins (STAT5a and STAT5b) is one of the earliest signaling events mediated by IL 2 family cytokines. |
Function: |
Carries out a dual |
Function: |
signal transduction and activation of transcription. Binds to the GAS element and activates PRL-induced transcription. |
Subunit: |
Forms a homodimer or a heterodimer with a related family member. Binds NR3C1 (By similarity). Interacts with NCOA1 and SOCS7. |
Subcellular Location: |
Cytoplasm. Nucleus. Note=Translocated into the nucleus in response to phosphorylation. |
Ptm: |
Tyrosine phosphorylated in response to IL-2 IL-3 IL-7 IL-15 GM-CSF growth hormone prolactin erythropoietin and thrombopoietin. Tyrosine phosphorylation is required for DNA-binding activity and dimerization. Serine phosphorylation is also required for maximal transcriptional activity (By similarity). |
Similarity: |
Belongs to the transcription factor STAT family. |
Similarity: |
Contains 1 SH2 domain. |
