Immunogen:
C57BL/10 mouse splenic T cells and concanavaline A-activated C57BL/10 splenocytes1
Characterization:
To insure acceptable performance each batch of product is tested by flow cytometry to conform to characteristics of a standard reference reagent.
Specificity:
M subunit (Mr 170 kDa) of the CD11b/CD18 heterodimerCD11b the integrin M subunit combines with CD18 the integrin beta2 subunit to form the integrin Mac-1 also known as complement receptor 3 (CR3) which mediates adhesion to C3bi and ICAM-1/CD54. 2 3 Mac-1 is expressed at varying levels on granulocytes macrophages dendritic cells NK cells and B-1 cells in the peritoneal and pleural cavities. 2-8 Mac-1 expression is rapidly up-regulated on neutrophils after activation and in the same time period that CD62L is shed from the cell surface. In addition to its role in binding C3bi on opsonized targets and mediation of the subsequent ingestion process Mac-1 is important as an adhesion molecule in the transendothelial migration of monocytes and neutrophils. 4 9 10 Cross-reaction of M1/70 to human CD11b has been observed on monocytes polymorphonuclear leukocytes and NK cells. 4 8
Working Dilution:
Flow Cytometry: 0. 3 ug/106 cells
Warning:
Reagents contain sodium azide. Sodium azide is very toxic if ingested or inhaled. Avoid contact with skin eyes or clothing. Wear eye or face protection when handling. If skin or eye contact occurs wash with copious amounts of water. If ingested or inhaled contact a physician immediately. Sodium azide yields toxic hydrazoic acid under acidic conditions. Dilute azide-containing compounds in running water before discarding to avoid accumulation of potentially explosive deposits in lead or copper plumbing.
Subcellular Location:
Membrane; Single-pass type I membrane protein (By similarity).
Similarity:
Belongs to the integrin alpha chain family. 1. Springer T. A. G. Galfre. D. S. Secher and C. Milstein. 1978. Eur. J. Immunol. 8:539. 2. Springer T. G. Galfre D. S. Secher and C. Milstein. 1979. Eur.
