Mouse Anti-Human CD22 (Clone MEM-01) mAb

In Western blotting this antibody reacts with non-reduced samples. The optimal working dilution should be tested by serial dilution.

100 ug in solution (PBS containing 15 mM sodium azide, pH 7.4). Precaution: Sodium azide is a poisonous and hazardous substance which should be handled by trained staff only.

In Western blot (e.g. Raji cell line) MEM-01 stained in the non-reduced sample only the upper band (ca 140 kDa) but not the weaker 130 kDa zone which was stained by several CD22 standards (these stained both zones). It was therefore assumed that the 140kDa zone corresponds to the so called CD22 beta (long) isoform. This assumption was not rigorously tested on transfectants expressing various alternative spliced forms of CD22. MEM-01 cross-blocked only one of the tested CD22 standards (OTH228, which was described as recognizing uniquely epitope “E” in the 4th workshop), it did not cross-block mAbs RFB-4, CLB22/1 and CLB-Bly1.
