Application Notes |
Western blotting (0.5-4 ug/ml), immunoprecipitation (20-40 ug/ml) and Immunohistochemistry (20-40 ug/ml). However, the optimal conditions should be determined individually. The antibody recognizes 42 kDa and 44 kDa of endogeneous Erk1/Erk2 (p44/p42 MAP kinase). |

Description |
The extracellular signal-regulated kinase 1 and 2 (Erk1 and Erk2) are closely related mitogen activated protein (MAP) kinases that are activated through the extracellular stimulation by many growth factors, mitogens and differentiation promoting agents via a protein kinase cascade. Erk1/2 are activated approximately 1000 fold by phosphorylation of neibouring threonine and tyrosine residues by Mek1 and Mek2. Both sites must be phosphorylated for maximum activity. Erk1/2 kinases are ubiquitously distributed in the nervous system, as well as a broad range of cells and tissues. These two kinases represent proximal kinases in the classical MAP kinase cascade pathway which links growth and differentiation signals at the cell surface (through tyrosine kinase) with transcription in the nucleus. |
Formulation |
100 ug (0.5 mg/ml) affinity purified, rabbit anti-Erk1/2 polyclonal antibody in phosphate buffered saline (PBS), pH 7.2, containing 50% glycerol, 1% BSA, 0.02% thiomersal. Precaution: Thiomersal is a poisonous and hazardous substance which should be handled by trained staff only. |
Immunogen |
Synthetic peptide derived from rat p42 MAP kinase |
Purification |
Affinity purified |
Storage and Stability |
The lyophilized antibody is stable for at least 2 years from date of receipt at -20C. The reconstituted antibody is stable for at least 2 weeks at 2-4C. Frozen aliquots are stable for at least 6 months when stored at -20C. Avoid repeated freeze-thaw cycles. |
Specificity |
The antibody recognizes 42 kDa and 44 kDa of endogeneous Erk1/Erk2 (p44/p42 MAP kinase). |
