Anti-HA (influenza hemagglutinin) polyclonal antibodies were generated in chickens against a synthetic peptide containing the influenza hemaglutinin epitope (YPYDVPDYA) conjugated to keyhole limpet hemocyanin (KLH). This is the same epitope as recognized by the HA.11 mouse monoclonal antibody. Eggs were collected from hyperimmunized hens, and the IgY fraction was isolated using a proprietary method. Epitope-specific antibodies were affinity purified over a peptide column, dialyzed against PBS, and the concentration was adjusted to 0.1 mg/ml. Sodium azide was added to a concentration of 0.02% (w/v), and 1.0 ml aliquots prepared. |
Quality Control |
Antibodies were analyzed using ELISA and western blots using an N-terminal HA-tagged protein. Secondary antibodies used in these quality control assays were alkaline phosphatase- (1:1000 dilution) and HRP-labeled (1:3000 dilution) goat anti-chicken IgY antibodies (Aves Labs), respectively. |

Chickens were immunized with a synthetic peptide containing the influenza hemagglutinin (HA) epitope (i.e., YPYDVPDYA) conjugated to keyhole limpet hemocyanin (KLH). After repeated injections, immune eggs were collected, the IgY fractions were purified from the yolks, and then affinity purified using a peptide column. The concentration of the eluate was then adjusted to 100 µg/mL, and the preparation was filter-sterilized. |
