Application info	The leukocyte count determination in seed fluid serves as a marker for the function of the accessory sexual glands. Leukocytes, especially polymorphic nuclear leukocytes (PML), are present in most human ejaculates. In normal microscopy, these can be confused morphologically with multinuclear spermatids. It is known that peroxidase enzymes are histochemically exclusively characteristic of the PMG granulocytes
Principle	Durch den Einsatz von Wasserstoffperoxid, lassen sich Peroxidase-positive Leukozyten (neutrophilen polymorphe Granulozyten) gelb bis braun färben. Andere Zellen (Spermien, Lymphozyten, Monozyten, Makrophagen und multinukleäre Spermatiden) bleiben ungefärbt (Peroxidasenegative). In diesem Test wird die Samenflüssigkeit mit den Reagenzien 1 und 2 behandelt, wobei sich lediglich Peroxidase-positive Zellen braun färben. Diese werden anschließend unter einem Phasenkontrastmikroskop identifiziert
