Formulation

This monoclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco's phosphate buffered saline (1X PBS) pH 7.2 - 7.3 containing 5.0% w/v trehalose with no calcium, magnesium or preservatives present.

Storage and Handling

The lyophilized antibody can be stored desiccated at -20°C to -70°C for up to twelve months. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid repeated freeze thaw cycles. No detectable loss of activity was observed after six months.

Other Applications Reported In Literature ?

Neutralization:Measured by its ability to neutralize IGFBP‑4 inhibition of IGF‑II-dependent proliferation in the MCF‑7 human breast cancer cell line. Karey, K. P. et al. (1988) Cancer Research 48:4083. The Neutralization Dose (ND50) is typically 10‑40 µg/mL in the presence of 0.3 µg/mL Recombinant Human IGFBP‑4 and 14 ng/mL Recombinant Human IGF‑II.

Each investigator should determine their own optimal working dilution for specific applications. See directions on lot specific datasheets, as information may periodically change.

Description

Specificity

Mouse Anti-Human Insulin-Like Growth Factor Binding Protein 4 (IGFBP-4) (Clone 82334) recognizes an epitope on Human IGFBP-4. This monoclonal antibody was purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype.

Background

Insulin-like growth factor binding protein 4, also known as IGFBP4, is a member of the insulin-like growth factor binding protein (IGFBP) family that are cysteine-rich proteins that act as a carrier protein. The protein binds both insulin-like growth factors (IGFs) I and II and circulates in the plasma in both glycosylated and non-glycosylated forms. Binding of this protein prolongs the half-life of the IGFs and alters their interaction with cell surface receptors. IGFBP-4 is a unique protein and it consistently inhibits several cancer cells in vivo and in vitro. The protein itself does not prevent the formation of cancer.1 However it may reduce the growth of cancer and act as an apoptotic factor.
