Formulation

This monoclonal antibody has been 0.2 µm filtered and lyophilized from modified Dulbecco's phosphate buffered saline (1X PBS) pH 7.2 - 7.3 containing 5.0% w/v trehalose with no calcium, magnesium or preservatives present.

Storage and Handling

The lyophilized antibody can be stored desiccated at -20°C to -70°C for up to twelve months. The reconstituted antibody can be stored for at least four weeks at 2-8°C. For long-term storage of the reconstituted antibody, aseptically aliquot into working volumes and store at -20°C to -70°C in a manual defrost freezer. Avoid repeated freeze thaw cycles. No detectable loss of activity was observed after six months.

ELISA Sandwich: This antibody is useful as the capture antibody in a sandwich ELISA. The suggested coating concentration is 2 µg/ml. A suitable detection antibody is PN:C1520 at a concentration of approximately 200 ng/ml. A suggested standard for this assay is PN:C359.

Neutralization: This antibody is useful for neutralization of Mouse CTACK bioactivity. The antibody dose required to neutralize 50% (ND50) of the biological activity of Mouse CTACK (at 1.0 ng/ml) is 2.5 - 10.0 µg/ml.

Rat Anti-Mouse Chemokine (C-C Motif) Ligand 27 (CCL27) (Clone 68623) recognizes an epitope on Mouse CCL27. This monoclonal antibody was purified using multi-step affinity chromatography methods such as Protein A or G depending on the species and isotype.

Background

Chemokine (C-C motif) ligand 27 (CCL27) is a small cytokine belonging to the CC chemokine family. It is associated with homing of memory T lymphocytes to the skin, and plays a role in T cell-mediated inflammation of the skin. CCL27 is expressed in numerous tissues, including gonads, thymus, placenta and skin. It elicits its chemotactic effects by binding to the chemokine receptor CCR10. CCL27 is one of the CC chemokines produced by epidermal keratinocytes and is suggested to be involved in the pathogenesis of inflammatory skin diseases. CCL27 modifies inflammation by attracting CCR10-positive and CCR4-positive cells into the lesional skin, and may participate in the pathogenesis of Th2-shifted skin diseases such as atopic dermatitis.
