Background |
Interferon-gamma (IFN-gamma) exerts its biologic effects by interacting with a single high affinity receptor, IFN-gamma receptor. This receptor is present on almost all cell types except mature erythrocytes. The IFN-gamma receptor consists of two polypeptide chains alpha and beta and requires association of JAK1, JAK2 and Stat1 for IFN-gamma signal transduction. In signal transduction, IFN-gamma induces tyrosine phosphorylation of IFN-gamma receptor alpha (IFNGR1) leading to the formation of a docking site on the activated receptor for Stat1, which specifically activates IFN-gamma induced gene transcription. The IFN-gamma receptor is structurally related to the recently cloned IL-10 receptor. GIR 208 binds IFNGR1 and blocks binding of IFN-gamma to the receptor. |
