Description: (+/-)-10-Hydroxycamptothecin, a naturally occuring indole alkaloid isolated from Camptotheca acuminate, is a cell-permeable and potent inhibitor of topoisomerase I. It has a broad spectrum of anticancer activity with selective inhibitory effect on the phosphorylation of histone H1 and H3, but less effect on other histones. (+/-)-10-Hydroxycamptothecin markedly inhibits the proliferation of Colo 205 cells in a dose-dependent manner. At the concentration of 5-20 nM, (+/-)-10-Hydroxycamptothecin arrests Colo 205 cells in the G2 phase of the cell cycle and triggers apoptosis through a caspase-3-dependent pathway.
