Description: Ocaperidone (R79598) is a potent and effective antipsychotic agent which acts as an antagonist of 5-HT2, dopamine D2 and a 5-HT1A, with Ki values of 0.14 nM, 0.46 nM, 0.75 nM, 1.6 nM and 5.4 nM for 5-HT2, a1-adrenergic receptor, dopamine D2, histamine H1 and a2-adrenergic receptor, respectively. Ocaperidone inhibits dopamine agonist (e.g. amphetamine or cocaine)-induced behavioral effects at low doses (0.014-0.042 mg/kg). It completely blocks the dopamine agonist induced behavior at slightly higher doses (e.g. 0.064 mg/kg). Ocaperidone shows pronounced haloperidol-like effects on the positive symptoms of schizophrenic patients but with risperidone-like low extrapyramidal (EPS) side effect liability and improved patient compliance.
