Epidermal Growth Factor

Der Epidermale Wachstumsfaktor (EGF) ist ein Protein, welches das Wachstum verschiedener epidermaler und epithelialer Gewebe stimuliert 1. Er besteht aus 53 Aminosäuren (6 kDa), welche über drei intramolekulare Disulfidbrückenbindungen verbunden sind 2. Er tritt verstärkt bei der Einleitung der Mitose auf. Zu Forschungszwecken kann er sowohl in vitro als auch in vivo verwendet werden. In machen Fibroblasten stimuliert er das beschriebene Wachstum auch in Zellkultur 1.

Epidermaler-Wachstumsfaktor-Rezeptor

EGF ist einer der wichtigsten Liganden der Epidermalen-Wachstumsfaktor-Rezeptorfamilie (EGFR). Der Rezeptor gehört zu Klasse der Transmembran-Glykoproteine und besteht aus einer extrazellulären Rezeptor-Domäne, einer transmembranen Region und einer intrazellulären Domäne, welche als Tyrosinkinase fungiert 3. Bindet z.B. EGF an EGFR, so findet eine Autophosphorylierung der Rezeptor Tyrosinkinase satt, was eine Aktivierung bestimmter Signaltransduktionswege zur Folge hat. Über diese Art der Signaltransduktion, werden unter anderem die Zellproliferation und die Zelldifferenzierung reguliert 4.

Forschungsbereiche

EGFR spielt eine besondere Rolle bei der Erforschung von Krebserkrankungen, da er in einer Vielzahl von Tumorzellen überexprimiert wird. Eine Überexprimierung von EGFR konnte besonders in soliden Tumoren wie z.B. Brustkrebs, Kopf- und Nackenkrebs, nicht-kleinzelliger Lungenkrebs (NSCLC), Nierenkrebs, Eierstockkrebs und Darmkrebs gezeigt werden 5. Die Aktivierung des Rezeptors wird ebenfalls mit der Resistenz gegenüber Chemotherapie und Bestrahlung in Verbindung gebracht.

Die beschriebenen Eigenschaften von EGFR machen den Rezeptor zu einem wichtigen therapeutischen Ziel bei der Krebsbehandlung, da Wechselwirkungen mit EGFR zu einer Unterbrechung der Signaltransduktion führen können 6. Als Inhibition von EGFR konnten besonders monoklonale Antikörper, welche gegen die extrazelluläre Rezeptordomäne gerichtet sind und niedermolekulare Verbindungen, welche die intrazelluläre EGFR-Tyronsinkinase-Aktivität stören, als effektive beschrieben werden 7. Sowohl monoklonale Antikörper als auch niedermolekulare Verbindungen können dazu führen, dass die Zellproliferation unterbrochen wird und die Apotose erleichtert ist. Folglich können den genannten Substanzen sowohl zytotoxische als auch zytostatische Eigenschaften zugesprochen werden 7.
